Establishment of Severe Traumatic Brain Closed Injury Animal Model in Non-anesthetic Mice

Abstract:Objective To establish an animal model of severe traumatic brain closed injury (closed TBI) in traffic or war in mice. Methods Severe closed TBI in mice was performed by a BIM-Ⅲ biomechanical machine with non-anesthesia. At the time points of 2h, 8h, 24h, 72h, 120h, 1w, 2w after injury, the neurobehavior of mice was measured by neurological examination and motor functional test. Besides, cerebral and lung fluid content, mortality rate, pathologic and ultrastructure picture were analyzed. Results 48h after TBI, the neurobehavioral and motor function scores decreased significantly than that in control group (P<0.05), the mortality rate was high, there were no significant differences in lung fluid content among dead mice in different groups, but the fluid content in injury group was significantly higher than that in control group (P<0.05), and brain injury such as cells tumefaction, necrosis were confirmed by pathologic and ultrastructure technique. Conclusion The imitation would be a valuable animal model for experimental investigation of severe TBI in accident of traffic or war.
