Advances of the Research on Isolation and Influencing Factors of the Domestic Preantral Follicles in vitro

In the mammalian ovary, about 99% of the follicles present in the cortex are in the form of a non-cavity. It is essential for isolating preantral follicles effectively. The separation of follicles is established in rodents. However, this remains a challenge in livestock which has constrained research. The separation methods for livestock animals preantral follicles can be largely divided into three categories: enzymatic digestion method, mechanical separation, combined enzyme-mechanical separation. According to the different tools researchers use, mechanical separation is generally divided into the following categories: cutting method, comb-scrape method, micromanipulation, and grinding. This article reviews the methodological history of isolating preantral follicles in vitro, focuses on the method for isolation of preantral follicles in vitro and influencing factors, such as animal species, age, processing time, and ovarian size and development, thus laying a theoretical foundation for establishing a more effective isolation method.
