Methods of Sample Preparation for Microdermatoglyphics Research of Squamate Reptiles

Abstract:
Squamate reptiles have a complicated shedding cycle and their epidermis have heterogeneous histological layers, but only the oberhautchen bears the repeating pattern of ornamentation. During the preparation of samples for scanning electron microscopy (SEM), the main source of variation in epidermal surface structure is the confusion of histological layers caused by unidentified stages in the shedding cycle. Using a method of gross dissection for examining preserved specimens to determine the stage in the shedding cycle, we can identify layers dispensing with histological techniques, and characterize their surface. A simple method for preparing the epidermis slice for optical microscopy is also introduced in this paper.
