An Ultra Performance Liquid Chromatography based method to quantity determine protocatechuic acid in ethanol extract of American cockroach

Abstract:Abstract: The present study reported an Ultra Performance Liquid Chromatography (UPLC) based method to quantity determine the protocatechuic acid in the ethanol extract of American cockroach. The column was washed with Waters ACQUITY UPLC HSS T3 column (2.1 × 100 mm; 1.8 μm) to option the salt solution (containing 0.1% formic acid, 25 mmol ammonium formate) as the mobile phase A, 100% acetonitrile as the mobile phase B at a flow rate of 0.5 mL· min-1. The specimen temperature was 25 ℃, and column temperature was 40 ℃, three detection wavelengths were set at 190~450 nm, 254 nm and 360 nm respectively. We detected the protocatechuic acid peak at a retention time of 3.3min with a base line separation. The protocatechuic acid showed a linear relationship (r> 0.99) in the linear range, and the average recovery ranged from 98.30% to 99.57% with the RSD <1%. The content of protocatechuic acid in the ethanol extract from American cockroach was quantity determined to be 51.38 μg/mL. The establishment of UPLC method to determine the content of protocatechuic acid will facilitate the quality control of American cockroach based productions in future.
